Uncontrolled behavior could be an important source of human error, especially in complex and hazardous situations where successful performance is critical. To study this, we tested the hypothesis that untrained and erratic responses to visual stimuli evoke implicit motor responses. In Experiment 1, 18 subjects were given 30 trials in a study phase, which consisted of responding as quickly and accurately as possible to squares presented at the center of a computer screen. Each trial consisted of 25 sequential trials during which an arrow appeared in one of four directions. In the test phase, the subjects participated in a speeded RT task, which consisted of comparing the speed of a moving arrow and that of the squares. In Experiment 2, the study and test phases were interchanged. In Experiment 3, the squares were replaced by two letters. The dosage and timing of expression of the transgene can be established by transfection of the gene into the target cells so that the gene is integrated in the genome of the cells and replicated with the host genome. This integration and expression can be achieved by any of a variety of techniques.
One such technique is, for example, the use of a retroviral expression system in which the therapeutic polynucleotide of the invention is initially cloned into a retroviral expression vector such as pMX-insulin.sup.1, which is subsequently transfected into a packaging cell line. A retroviral expression system is preferable to cell-based transfection of the polyisomcyin polymers since it provides for integration of the therapeutic polynucleotide in the target cells, thus resulting in stable expression. The expression of the therapeutic polynucleotide is easily controlled by the person of ordinary skill in the art by varying the amount of the therapeutic polynucleotide provided in the viral stock.
Once a cell is transfected with the retroviral polynucleotide of the invention, that cell may be further genetically modified to produce the polyisomcyin polymers. This may be accomplished by addition of various exogenous DNA to the target cells.
Specific techniques for accomplishing these modifications are described in detail herein. Once the preferred form of the invention has been developed, it will be recognized by those skilled in the art as suited for application to other polymers, such as human polyisomcyin, by routine experimentation.
In accordance with another embodiment of this invention, polyisomcyin is covalently attached to an agent which is capable of specifically delivering the polyisomcyin to various sites within the body in order to enhance the therapeutic effect.
